Assays to measure intracellular antiviral response:

The best point to measure antiviral response in cells is accessing the induction of anti-viral response pathway in the very beginning by measuring elevated synthesis (induction of mRNA and protein expression) of type I interferon (Interferon-alpha and Interferon-beta) and pro-inflammatory cytokines, such as TNF-alpha, IL2, IL-8, IL-6 and JAK1/2.

Commonly used methods to characterize induction of intracellular anti-viral response include real-time qRT-PCR method to access elevated mRNA expression levels of the following genes: Interferon-alpha, Interferon-beta, JAK1/2, IL8, and TNF-alpha. Pre-designed Taqman-based kits are commercially available.
